Mechanical Dedikaryotization: what a mouthful…but if you know…you know. The power of the monokaryon is something not to be underestimated and overlooked. The simplicity of a protocol written up by @dedikariotizationdaddy and derived from Hermilo Leal-Lara’s work on “monokaryotization” from the early 1980’s is the basis for this week’s episode. Outside of Julian’s work creating a monokaryotic library from fungal phenotypes, no other company or institution we are aware of has presented an interest in replicating the work of splitting all of their dikaryons to increase the amount of cultivators taking “crosses” into their own labs. Over this winter @mainecapnstemco will be developing a library of cryogenically preserved “mons” to be used in future breeding programs. These mons will be available through our website as we confirm the lack of clamps and start the lines and loops of cryo-preservation. As for today’s Deep Dive, we took extra care and time to delve into the realities of mechanical-dedi-work. While there are other methods for isolating the two “mons” that make up a “di”, we wanted to initially explore this simple methodology with our dedicated MycoWizards members to rekindle an interest in lab work beyond the expansions for fruit body production. So dust off your microscopes, bust out your blenders and let's get down to it!
